N-去甲基万古霉素单组份的分离与精制

【摘要】 本文研究了适用于分离万古霉素等大分子抗生素的离子交换葡聚糖凝胶梯度柱层析法,用此方法分析国内外万古霉素样品,发现国产“万古霉素”与国外样品的保留时间不同,相差148分,故两者不是同一物质。现经试验证明,国产“万古霉素”确非万古霉素,而是N-去甲基万古霉素。用此法并配合结晶的方法精制N-去甲基万古霉素精制品,经高效液相色谱法测定,纯度为99.05%。

【Abstract】 A gradient column chromatography packed with CM-Sephadex C-50 for separation and purification of components of domestic made Vancomycin was developed. The frac-tionation patterns of components were plotted, the main component was isolated and recrystallized. A single component was ob- tained and the purity was found to be 99.05 %. The retention time of the main component of the domestic made Vancomycin is different from that of Vancomycin but corre-seponding to that of N-demethyl Vancomycin.
