急性白血病外周血染色体脆性部位观察
FRAGILE SITES IN PATIENTS WITH ACUTE LEUKEMIA
【摘要】 本文介绍用低浓度小牛血清、低叶酸,较高pH值的培养方法,对10例白血病和10例正常人外周血染色体畸变率及脆性部位进行了观察。结果表明:病人组染色体结构畸变率明显高于对照组;G显带定位48个断裂点,其中脆性部位31个:与癌基因对位者7个,位于癌基因旁2个。并简要讨论了脆性部位在白血病中的可能作用。
【Abstract】 PHA-stimulated peripheral blood white celts were incubated in 199 medium supplemented with 5% calf serum at 37℃ for 96 hours. In order to reveal more chromosome fragile sites the pH of culture medium was set at 7.5. G-banded chromosome analyses were carried out after microphotography of fragile sites in order to specify their precise localization. Ten patients aged from 4 to 28 years were studied. Three of these patients were with ANLL, five ALLand 2 LCL. Ten normal controls were also examined. The results were: (1)the chromosome structural aberration rates of the patients(12.46%)were much higher thanthose of the control group(0.8%) P<0.001), (2)G-banding showed 48 breakpoints, 15 of them were fragile sites, 7 were at the same baud as oncogenes, 2 were near the band as fragile sites or oncogenes, (3)six patients carried multiple fragile sites and in the control group, none carried any fragile sites. The possible role of the fragile sites in acute leukemia was briefly discussed.
