β-内啡肽、促肾上腺皮质激素、5-羟色胺和去甲肾上腺素对疼痛和针刺镇痛的影响
EFFECTS OF BETA-ENDORPHIN, ACTH, 5-HYDROXYTRYPTAMINE AND NOREPI-NEPHRINE ON PAIN AND ACUPUNCTURE ANALGESiA IN RATS
【摘要】 本工作采用化学毁损弓状核及摘除大鼠垂体、荧光分析及放射免疫测定等方法,观察到: (1)毁损弓状核后脑内β-内啡肽含量降低67%,针刺镇痛效应明显下降:将此种处理大鼠的垂体切除后,针效几乎消失,同时,电针后脑去甲肾上腺素含量明显高于电针对照组。 (2)促肾上腺皮质激素不仅提高基础痛阈,增强针刺镇痛效应和吗啡镇痛效应,同时也加强中枢5-羟色胺能神经元系统的功能。 (3)大鼠摘除垂体后,针效明显减弱,脑内5-羟色胺及其转换率均下降,补充ACTH后,针效基本恢复,脑内5-羟色胺含量及其转换率也显著上升。
【Abstract】 In the present study, we examined the effects of lesion of the hypothal-amic arcuate region after neonatal treatment with monosodium glutamate(MSG) on the content of /?-endorphin, 5-hydroxytryptamine (5-HT) and norepinephrine (NE) in the brain and acupuncture analgesia in rats.In the rats treated neonatally with MSG the brain content of /?-endorphin by 67%, as compared with those of the control animals. The effect of acupuncture analgesia was significantly weakened. The content of NE after ele~ ctroacupuncture was relatively higher in the MSG treated group than in the control group. After being treated with MSG the rats were hypophysectomi-zed, and the analgesic effects of acupuncture were then nearly completely abolished. The content of NE after electroacupuncture in the hypophysecto" mized group was significantly higher than that of rats treated only with MSG.In hypophysectomized rats the brain 5-hydroxyindoleacetic acid (5-HI-AA) and total amount of 5-HT increased markedly. However, the acupuncture analgesic effect was much less than that of the sham operation group. There was no marked increase in the brain 5~HT leval and no marked decrease in the brain NE after acupuncture. After hypophysectomized rats were given ACTH intraperitoneally, the brain 5-HT leval and Its turnover rate increased markedly, and the recovery of acupuncture analgesic effect ensued.Injection intramuscular of ACTH also increased the acupuncture and morphine analgesic effect, the total amount of the brain 5-HT and its turnover rate significantly, but there was no marked change in the brain /9-endorphin leval.
