NHG-1人脑胶质瘤裸小鼠模型特征的观察
Observation on Characteristics of Nude Mice-Human Glioma Model-1(NHG-1)
【摘要】 本实验将人脑恶性胶质瘤体外细胞系SHG-44接种于NC裸小鼠,初代成功率为7/11,4年内33代系统传代移植成功率均为100%。肿瘤倍增时间7天,细胞周期时间2.5天,宿主生存时间54±15天。肿瘤组织非常容易向周围软组织浸润性生长,通过光镜、电镜的形态观察,LDH同功酶测定,PAP免疫组化标记CFAP和染色体核型分析,证明移植瘤符合人脑恶性胶质瘤的特征。我们还研究了抗癌药ACNU和BCNU对NHG-1的治疗作用。与此同时,对从蜂王浆中提取的10-羟基癸烯酸的作用也进行了评价。
【Abstract】 Human glioma cell line SHG-44 transplanted into NC nude mice, the initial transplantability was 7/11,and the transplantability on subcequeent serial transplantation of 33 passages in 4 years was 100%. The transplantable tumors potential doubling time was 7 days and cell cycle time was 2. 5 days. As for survival time of host,the averagelife span±standardized deviation was 54±15 days. The transplantable tumors were subcutaneous masses with a variable tendency to invade surrounding soft tissues. By light and electro microscopies for morphological observation, LDH isozyme assay, PAP immuno-peroxidase labelling GFAP and chromosome analysis, transplantable tumors were conformed to the characteristics of human malignant glioma. We also studied the therapeutic of anticancer drugs such as ACNU and BCNU in the NHG-1 model and at the same time the effect of 10-hydroxy-2-decenoic acid from royal jelly was alco evaluted.
