竹红菌甲素对红细胞膜光敏作用机理的研究
THE MECHANISM STUDY OF HYPOCRELLIN A PHOTOSENSITIZATION IN ERYTHROCYTE MEMBRANE
【摘要】 本文研究了甲素对红细胞膜的光敏化作用,证实了此光敏作用是需要氧分子参加的。还使用单重态氧1O2猝灭剂NaN3和DABCO,以及O2-超氧阴离子自由基猝灭剂SOD酶,来研究甲素对红细胞膜蛋白和膜类脂光氧化过程的反应机理。结果表明,NaN3、DABCO和SOD酶分别对膜蛋白SH基和膜类脂的光氧化没有明显的影响,仅NaN3对膜蛋白光敏交联有部分抑制作用。而这些光敏作用在D2O中也未见明显加剧。说明甲素对红细胞膜的光氧化机理可能涉及1O2或O2-,但也有其他的“活泼态氧”在起作用。
【Abstract】 In this paper, the effects of hypocrellin A on photosensitizalion of erythro-cyte membranes have been investigated. It has been elucidated that photosen-sitization takes place in the presence of oxygen. The singlet oxygen(1O2) quenchers: NaN3 and DABCO, and superoxide anion radical (O2-) quencher:super-oxide dismutase, were used to examine the mechanism of hypocrellin A-sensi-tized cross-linking of erythrocyte membrane proteins and photooxidation of unsaturated lipids.The results show that NaN3, DABCO and superoxide dismutase do not cause significant effect on the photooxidation of SH group in protein and unsaturated lipids, but NaN3 can partially inhibit photosensitized cross-linking of membrane proteins.The lifetime of 1O2 in D2O and H2O are 30μs and 3μs respectivelyo If 1O2 would play an important role in hypocrellin A-sensitized photodynamic effects on erythrocyte membrane, then photosensitization would significantly increase in the DaOo But in our experiments, D2O effect was not observed. It is suggested that the photodynamic effect of 1O2 and O2- unlikely plays an important role, but other forms of activated oxygen may take part in photooxidation.
