微波照射家兔睾丸抑制精子生成的有效温度阈值

【摘要】 利用微波的热效应原理,对实验动物睾丸进行局部照射,以提高睾丸内部温度,改变精子生成的物理环境,从而抑制精子产生,达到节育目的。本文在相同的实验照射条件,和用睾丸中心与阴囊表面同时进行对比性测温的情况下,对实验规定的不同温度组动物的实验结果进行比较,目的在于探讨微波用于节育时的有效温度阈值。实验用微波源为WL-200型医用微波理疗机。动物用体重2.5公斤以上、照前精子密度在2亿/毫升以上的雄性家兔共60只,随机分为4个大组。各组实验规定温度是以微波照射后,睾丸中心温度上升至实验额定温度为准。4个实验组照射时睾内温度分别确定为:41℃,43℃,44℃,46℃。各组动物照后定期分别进行精子密度动态观察和睾丸组织切片检查。实验结果表明,经微波一次照射20分钟后,睾丸内温度升至41℃时,受照动物精子密度并不下降,为节育无效温度。睾内温度43℃组中一半动物精子密度明显下降,但维持时间较短,节育效果不理想。睾内温度44℃组,在20分钟照射下,无论照射1次或2次,所有实验动物精子密度均有明显下降,于照射后15—19天即可降低至0.3亿/毫升以下,维持低水平时间一次照射亚组在23天左右,两次照射亚组约37天。本组在实验结束时,平均精子密度及睾丸组织学改变均能较好恢复,属于节育有效温度。睾内温度46℃组

【Abstract】 Making use of the thermal effective tenet of microwave, we applied local irraditation to experimentalanimal testis to raise the temperature of the interior of the testis, to change the physical environment of the testicular growth, and put the testicular development under control in order to obtain contraceptive effect. The measured temperature of contrast between the testicular cone and the surface of the scrotum were done under identical conditions of the experimental irradiation. Comparison of the results of the groups at different temperature was done to find out the effective temperature threshold for contraception by microwave. The microwave which is applied in the experiment is medical microwave physiatries equipment of WL-200 type, the animals all weighed above 2.5Kg. the 60 male rabbits used had sperm density above 2 hundred millions/ml, before the irradiation and were divided into four groups. The temperature of the testicular cone of each group after irradiation was set for 41℃, 43℃, 44℃, 46℃ respectively. After irradiation, regular observation of the development of the sperm density and check-up of the testicular tissue section were done. Results showed that the temperature of the testicular interior rose to 41℃, after being irradiated once for 20 minutes, without decline in the sperm density of the animal. In the group with interior temperature of 43℃, half of the animals showed a marked drop in sperm density, but the effect lasted only a short time and not acceptable for contraception. In the group with 44℃, all the animals showed marked drops of sperm density under 20 minutes irradiation. no matter under irradiation for once or twice. The sperm counts reduced to less than 0.3 million/ml from 15 to 19 days after the irradiations. To maintain this low level for 37 days, two irradiations had to be given. After this, the animals in this group gradually recovered normal spermatogenesis and normal histology of the testis, showing that this is probably the optimal temperature threshold for effective reversible contraception. In the group with 46℃, most of the animals presented edematous skin of the scrotum after the second irradiation. A few manifested variable burn lesions, so it can not be used as a contraceptive measure.
