重组γ干扰素对人外周血粘附细胞DR抗原的调节作用

【摘要】 用抗HLA-DR抗体介导的补体依赖51Cr释放细胞杀伤抑制试验,从人外周血粘附细胞和HLA-DR+细胞株的细胞培养上清液中检测到HLA-DR抗原。进而,发现γ干扰素在体外不仅维持和增加粘附细胞表面的DR抗原,而且加快DR抗原从细胞表面的脱落,提示γ干扰素通过增强DR抗原的合成来维持和增加细胞表面的DR抗原。

【Abstract】 A reproducible and sensitive antibody mediated complement dependent cytotoxicity inhibition assay has been developed for the detection of cell-free HLA-DR antigen. Human peripheral blood adherent cells (PBAO) as well as DR+ monocytes and B lymphoblastoid cell lines but not T lymphocytes were observed to shed DE antigen in vitro. Human recombinant intrferon-γ(IFNγ) induces the expression of DR antigen by PBAO by increasing both the number of cells expressing DE antigen and its density per cell. After incubation with IFN γ, PBAO also shed significantly more DR antigen. The degree of DE shedding is dependent on the dose of IFN γ. Results demonstrated that increased cellular HLA-DR expression induced by IFN γ was accompanied by increased shedding of DE antigen into the culture supernatants.
