氯丙烯对周围神经纤维超微结构的影响

【摘要】 本实验观察到氯丙烯对有髓和无髓神经纤维均具毒害作用,损害部位呈局灶性分布,表明对神经纤维直接发生作用。神经纤维的改变主要是轴突髓质内微管和神经微丝形态明显变性、数量显著减少,这种改变迅速且严重,而线粒体变化较轻和缓慢。微管和神经微丝变性和数量减少将可能影响或破坏轴浆运输。

【Abstract】 Abstract The observation on the number of microtubule and neurofilaments in myelinated nerve fibers (diameter:3~5μ) of normal rat and the rat exposed to allyl chloride (dosage: 150mg/kg) for 3 to 7 times was reported.The main values are:110±28.3/10μ~2 and 1063±228.2/10μ~2., 88±30.7/10μ2 and 969±306.6/ 10μ~2 73±28.8/10μ~2 and 643±390.8/10μ~2. Although there was no significant decrease in the number of microtubuless exposed for 3 times (P>0.05), the decrease in number of neurofilaments is significant (0.05>P>0.01). After 7 times of exposure the number of both fine structures decreased markedly (P<0.01). The degeneration of microtubules and neurofilaments constituted the chief ultrastructural change in myelinated and unmyelinated nerve fibers. There revealed slight degeneration of mitochondria and no degenerational change in myelin sheath. The lesion area showed focal distribution, and the thick fibers appeared to be more seriously damaged in comparision with the thin ones,, Our results suggest that allyl chloride exerts direct effects on axon and the pathological changes of axons are not the consequence secondary to changes of cell body. Nevertheless, neurofilaments and microtubules seemed to be more sensitive than mitochondria and the lesion could be considered as axonal degenerative neurophage.
