石菖蒲主要成分α-细辛醚致突变研究

【摘要】 以三种短期试验检测α-细辛醚的致突变性。结果表明α-细辛醚对鼠伤寒沙门氏菌TA98有致突作用。染色体畸变分析试验,α-细辛醚在剂量为185.2mg/kg时大鼠骨髓染色体畸变率为3.8%,与对照组0.3%相比畸变率显著上升(P<0.01),提示α-细辛醚对染色体有断裂效应。小鼠骨髓微核试验表明,试验组嗜多染红细胞的微核百分数虽有轻度上升,但与对照组相比无统计学意义,而环磷酰胺微核数则显著升高。

【Abstract】 Mutagenicity of α-asarone wasstudied by three short-term tests. TheSalmonella/microsomal test system wasused for the detection of reverse muta-tion. The results showed that α-asaroneexhibited mutagenicity towards strainTA98. The clastogenic potential wasevalnated dy direct cytogenetic analysisof mataphase and micronucleus test. Ata dosage of 185.2mg/kg, the chromosomalaberration rate in the bone marrow ofrats was 3.8%, which was significant(p<0.01) in comparison with the controlgroup (0.3%), suggesting the clastogeniceffect of α-asarone. Micronucleus test inthe bone marrow of mice indicated thatthe percentage of polychromatic erythro- cytes with micronuclei in exposed groupswas increased slightly, but no significantdifference existed in comparison with thecontrol. A significant increase in numberof micronuclei was observed in animalstreated with cyclophosphamide.
