马桑内酯对麻醉狗脑脊液中单胺类神经递质代谢物含量影响的初步探讨
A Preliminary Investigation on Effects of Coriaria Lactone on Metabolites of CNS Transmitters in CSF of Anestheized Doge
【摘要】 作者通过狗口服丙磺舒阻断酸性物质向外周血循环转运的方法来测定肌注马桑内酯1mg/kg,前、后脑脊液中去甲肾上腺素、多巴胺和5-羟色胺的代谢物3-甲氧-4羟苯一乙二醇、高香草酸和5-羟吲哚乙酸的含量改变,以了解三者的更新率。初步结果表明:高香草酸的含量显著下降(P<0.05),5-羟吲哚乙酸的含量有所下降(P>0.05)但无统计学意义,3-甲氧-4羟苯一乙二醇影响不大。
【Abstract】 Coriaria lactone is proved to be an effective drug for schizophr enia by clinical investigations in our university hospital,whereas it provoked typical epilepsy EEG in experiments with rabbits. It is not clear whether these effects are related to CNS transmitters. For the difficulty of directly measuring CNS transmitters,the authers determined the metabolites of NA,DA and 5-HTin CSF of dogs before and after the intr amuscular administration of coriaria lactone (1mg/kg) while the transportation of these metabolites to peripheral circulation was blocked with oral probenecid. Sephadex G10 columns were used to seperateMHPG, HVA and 5-HIAA in the CSF and their contents were determined by spectrofluorophotometry RF-510. The results showed that coriaria lactone markedly decreased the amount of HVA (P<0.05) but not that of MHPG and 5-HIAA.
