感染杜氏利什曼原虫DBA/1小鼠的非特异性免疫抑制机理初步探讨

【摘要】 用~3H—TdR 掺入法,对感染了杜氏利什曼原虫中国株的DBA/1 小鼠的脾细胞在有丝分裂原诱导下的淋转反应进行了观察。受染小鼠脾细胞对 Con-A、LPS和PHA的淋转反应明显受抑,去除其中尼龙丝柱粘附细胞后,淋转反应恢复正常;此粘附细胞亦可抑制正常DBA/1小鼠脾细胞对上述有丝分裂原的淋转反应。消炎痛可部分解除此粘附细胞对淋转的抑制,说明受染小鼠淋转反应低下系此粘附细胞抑制所致,其抑制功能可能与前列腺素有一定关系。

【Abstract】 In vitro proliferative responses to mito-gens (Con-A, LPS and PHA) of the spleencells from DBA/1 mouse infected by Leish-mania donovani were markedly inhibited,as showed by the measurements of tritiatedthymidine incorporation, and the res-ponses reversed to normal level whenadherent cells were depleted from themouse spleen cells by passing through any1on wool column. In mixed cultures, theadherent cells were capable of suppressingthe responses of normal DBA/1 spleen cellsto those mitogens as well. Indomethacin,when added to the culture, was found topartly abrogate the suppression capacity ofthe adherent cells. These observations. sug-gest that the non-specific immune suppres-sion in visceral leishmaniasis may be mediatedby the adherent cells whose immune sup-pressing function is partly associated withthe synthesis of prostaglandin. Characteriza-tion of T, B and Mφ by anti-microcytotoxi-city test,IFA (mIg) and Nonspecific EsteraseStain respectively in a preliminary studyshowed an increased high number of Mφ inthe adherent cells, indicating the Mφ as thesuppressing calls.
