四氧嘧啶所致的糖尿病对大鼠CFU-GM产率的影响
THE EFFECT OF ALLOXAN INDUCED DIABETES ON CFU-GM OF MICE
【摘要】 本文观察到四氧嘧啶所致的糖尿病小鼠CFU-GM 产率降低,胰岛素治疗可使之部分恢复。胰岛素能直接提高扩散盒及体外培养中CFU-GM 产率,其在体外的作用与浓度有一定关系。在一定范围内高葡萄糖浓度可促进体外培养中CFU-GM 产率。
【Abstract】 The experimental diabetic mice had high blood glucose and low insulin concentration produced by a single dose of alloxan 75mg/kg intravenously. No obvious toxic effects of alloxan on liver and kidney were discovered by histological examination. In the experiment of the effect of diabetes on the production rate of GFU-GM, the numbers of CFU-GM in diffusion chambers with alloxan were not less than that of control group (p>0.05), but the numbers of CFU-GM in diffusion chambers of diabetic receptor mice werw less than that of control group (p<0.01) and of insulin-treated group(p<0.01). In pairing experiment, the numbers of CFU-GM in the chambers with insulin increased(p<0. 001). Insulin with the concentration of 10-9 unit/ml medium promoted the production rate of CUF-GM in vitro (p<0.01). When the concentration of insulin increased from 10-8 to10-1 unit/ml, a little increase in the production rates of CUF-GM was observed, but without any statistical significance. When glucose was directly added to oulture dish, the numbers of CFU-GM increased correspondingly as the concentration of glucose increased from73.5 to 273.5 and 473.5mg/dl culture medium (P<0.01).
