多能造血干细胞(CFU-S)的不均一性——Ⅰ.9天CFU-S和13天CFU-S的某些性质比较

【摘要】 与9天CFU-S 比较,13天CFU-S 产生较多的脾集落数,自我更新能力较强,对放射的敏感性较低。9天CFU-S生成的脾集落细胞类型以单系主要是红系细胞为主;13天CFU-S生成的脾集落是以混合系主要是红、粒和巨核三系细胞混合为主,表明13天CFU-S是一类更为原始的多能造血干细胞。

【Abstract】 The properties of hematopoietic stem cells generating spleen colonies (CFU-S) Were studied. It was found that. 1) the number of spleen colonies harvested on the 13th day after transplantation of mouse bone marrow cells into lethally irradiated recipient mice was 1.6 times that of the 9th day; 2)the self-renewal capacity of day-13 CFU-S was significantly higher than that of day-9 CFU-S; 3)the radiation sensitivity of day 13 CFU-S was significantly lower than that of day 9 CFU-S With Do Values of 1.47 ± 0.03 and 1.04 ± 0.05 Cry respectively. It was also shown that on the 9 th day, single hematopoietic li- neage especially erythroid colonies in the spleen were of predominant type,however, the mixed colonies (especially erythroid,granulocytic and megakaryocytic mixed colonies) became predominant by the 13th day. It was confirmed that CFU-s is a heterogeuous population. It is indicated that day 9 CFU-S and day13 CFU-s belong to two differentsubgroups aud day13 CFU-S are more primitive pluripotent stem cells.
