老年小鼠的造血机能

【摘要】 老年小鼠骨髓中CFU-S相对数量少于年青小鼠,而 CFU-S总数则两者相等。移植等量来源于老年或年青鼠的 CFU-S后,受体鼠股骨中有核细胞相等,但在脾脏中的有核细胞以来源于年青供体者多些。以同样供体骨髓细胞输入不同年龄受体鼠后,老年受体鼠脾结节数明显少于年青小鼠。以不同年龄组供体鼠 CFU-S输注给不同年龄组受体鼠后,生成的脾结节数经统计学处理,提示造血干细胞与造血微环境均遭老化过程的影响。

【Abstract】 The number of CFU-S per 5×104 bone marrow cells is much less in old than in young mice, but the total number of CFU-S per femur is equal between old and young mice as estimated by spleencolony technique of Till with McCulloch. The total number of nucleated cells found in femur bone marrow of lethally irradiated recipient mice after transplanting equal number of CFU-S from young and old mice showed no significant difference but the number of nucleated cells found in spleen or recipient mice after transplantation was more in those receiving cells from young donors than from old mice. The number of spleen-colonies generated from 5x10* donor bone marrow cells was significantly higher in young recipient mice than in old ones while the same batch of bone marrow cells from young donors was transplanted. while the bone marrow cells from donors of different ages were transplanted into recipients of different ages, the number of spleen colonies harvested was the highest in young recipients receiving cells from young donors and the least in old recipients receiving cells from old donors. The other two groups were intermediated between these two groups. These results suggested that both hemopoietic stem cells and hemo-poietic induction microenvironment undergo senile changes but adequate methods to demonstrate these ohanges need to be further explored.
