甲醇毒理学研究:Ⅱ.甲醇代谢及其酶动力学观察
TOXICOLOGCAL STUDIES OF METHANOL, II THE METABOLISM OF METHANOL AND ITS ENZYMATIC KINETICS OBSERVATION
【摘要】 本研究表明:无论有无NADPH的存在,苯巴比妥钠预处理均可使大鼠肝匀浆中甲醇代谢率明显增加,而SKF—525—A预处理则无效。在体外测试系统中加入叠氮化钠可明显抑制甲醇代谢,而加入NADPH则可增加其代谢。相反,SKF—525—A在这方面无任何作用。 甲醇代谢的酶动力学研究表明:Km和Vmax分别为4.4mM和1.1nM/mg蛋白/分。当反应体系中加入2.5mM和12.5mM乙醇时,Km值分别变为6.7mM和11.4mM,而Vmax不变。然而,当加入1.0mM叠氮化钠时,Vmax减少而Km增加。
【Abstract】 The metabolic rates of MtOH in liver homogenate (LH) of rats pretreated with sod. Phenobarbital with or without the preseace of NADPH were increased significantly while SKF 525-A pretreatment had no effect, sodium azide significantly inhibited and NADPH increased the MtOH metabolism when they were added to the in vitro assay system.In contrast,SKF 525-A did not show any effect in this respect.The studies of enzymatic kinetics on MtOH metabolism showed that the Km and Vmax were 4.4 mM and 1.1 nM/mg protein/ruin, respectively. When 2.5 mM and 12.5 mM ethanol were added into the system, the Km changed into 6.7 mM and 11.4 mM, respectively, with Vmax unchanged. However, Vmax decreased whereas Km increased when 1.0 mM Sod. azide was added.
