~3H对胚胎猪肝胸腺嘧啶核苷酸合酶的动力学效应
Tritium Kinetic Isotope Effect on Thymidylate Synthase from Fetal Pig Liver
【摘要】 胸腺嘧啶核苷酸合酶(Thymidylate synthase)的催化作用通过有序机制(Ordered sequential mechanism)进行。在不同浓度的蝶酰单谷氨酸衍生物(CH2-H4PteGlu1)存在时,测定与(5-3H)dUMP的消耗相关的V/K效应,並通过一偶联体系,测定与(5-3H)dUMP和(2-14C)dUMP混合物的消耗相关的V/K效应.结果表明,3H对胚胎猪肝胸腺嘧啶核苷酸合酶的动力学效应,与以相同底物所进行的产物抑制作用和死端抑制作用而确立的底物加入和产物释出的次序相符。
【Abstract】 Thymidylate synthase catalysis proceed via an ordered sequential mechanism. The results of studies on the kinetic isotope effects on V/K of thymidylate synthase purified from fetal pig liver associated with consumption of (5-3H) dUMP and associated with the consumption of a mixture of(5-3H)dUMP and ( 2-14C ) dUMP in a coupled assay system are consistent with the conclusions about the order of addition of substrates and of release of products determined by product and dead-end inhibition studies with the monoglutamate substrate, CH2-H4PteGlu1.
