高温效应在血卟啉光敏疗法中的协同作用
A STUDY ON THE SYNERGISTIC EFFECT OF TEMPERATURE INCREASE IN HPD PHOTODYNAMIC THERAPY OF CANCER
【摘要】 本文对血卟啉光敏疗法治疗恶性肿瘤中与治疗效果有关的几个因素——光功率、光剂量、温度——进行了细胞和动物实验研究。结果表明,血卟啉光敏反应的程度和对癌细胞的杀伤效果取决于辐照的光剂量而不是光功率,这与光敏反应的机理相符。但在实际治疗中,若采用较大的光辐照功率,会引起肿瘤内的温升,当超过41.5℃的阈值温度后,高温与光敏反应有杀伤癌细胞的协同作用。根据动物实验的结果,在应用血卟啉光敏疗法治疗恶性肿瘤中,采用200~300mW/cm~2光功率密度的光辐照,最能利用高温的协同作用,达到好的效果。
【Abstract】 In the treatment of cancer by photodynamic therapy using hematoporphyrin derivatives, the problem on whether their therapeutic efficiency is dependent on the energy of irradiation or on radiation power and how the effect of intratumor temperature increase contributes during the process had been raised.In an in vitro experiment on a human hepatoma cell line we found that the cell survival was dependent on the dose of irradiation and not on radiation power, which agreed with the photodynamic reaction mechanism. On the other hand, our results on the sarcoma-bearing kunming albino mice showed that the effectiveness of HPD plus ligh treatment depended on radiation power. We had measured the temperature increase in mouse tumors treated with light of same dosage but of different optical power density and made comparison by histopathology. The results show that the intratumor temperature increase induced by radiation of higher optical power density had a synergistic effect on damaging tumor tissues.And the optical power density to be recommended was range of 200-300mW/cm~2.
