血红蛋白种间分子杂交以及树鼩进化地位的探讨
THE EVOLUTIONARY RELATIONSHIP OF TUPAI AS VIEWED FROM THE INTERSPECIFIC MOLECULAR HYBRIDIZATION OF HEMOGLOBIN
【摘要】 为了探讨树鼩(Tupaia belangeri chinensis)的分类地位及其与灵长类动物的亲缘关系,本文利用血红蛋白种间分子杂交技术,以人类血红蛋白HbA为标准,分析比较了树鼩血红蛋白与原始灵长类动物懒猴(Nycticebus coucang)血红蛋白的结构异同。在碱性条件下,三种血红蛋白的电泳迁移率大小为:人>懒猴>树鼩。树鼩Hb(α2Tup和β2Tup)和HbA(α2Aβ2A)杂交可以产生两种杂种分子 (α2Tupβ2A和α2Aβ2Tup)。HbA和懒猴Hb (α2Nycβ2Nyc) 杂交也可产生两种杂种分子(α2Nycβ2A和α2Aβ2Nyc)。结果表明,树鼩Hb与懒猴Hb的α链净电荷相似。认为树鼩与懒猴的分类地位相近。
【Abstract】 The evolutionary relationship of tupai (Tupaia belangeri chinensis) was studied through interspecific molecular hybridization of the hemoglobins between tupai and human, tupai and loris (Nycticebus coucang) as well as human and loris. The Rf values of electrophoresis of the Hb in pH 8 . 6 were found to be as follows: human>loris>tupai. The results of interspecific molecular hybridization between tupai Hb (α2Tupβ2Tup) and human adult hemoglobin (HbA) (α2Aβ2A) showed that, besides the two Hb molecules just mentioned, two other Hb molecules, the hybrid Hb molecules α2Tupβ2A and α2Aβ2Tup, were found. The hybridization between HbA and the Hb of lorts (α2Nvcβ2Nc) could also form two hybrid Hb molecules, α2Nvcβ2A and α2Aβ2Nvc. These results suggest that the α-chain of tupai is similar to that of loris Hb in net electric charge and hence substantiate the viewpoint that tupai may be considered as one of the primitive primate species.
