用凹孔板培养-压印法对鼠疫菌初期集落的观察
OBSERVTIONS ON PRIMARY COLONIES OF YERSINIA PESTIS WITH HOLLOW SLIDE CULTIVATION-IMPRESSION METHOD
【摘要】 本文介绍了凹孔板培养-压印法的操作步骤和方法。鼠疫杆菌在0.025%亚硫酸钠加1%溶血凹孔板(?)脂培养基上培养2~6小时,其压印标本集落的恃征为:由增大的两极浓染的菌体组成长短不等的菌链。菌链自然弯曲如“绳头状”、“蚯蚓状”、“绳结状”;同时由数量不等的菌链丛集成“束状”、“指印状”、“斗笠状”及不规则盘绕的菌族集落。散在分布。组成菌链的菌数和构成集落的链数,随培养时间的延长和接种菌量的加大而增加。出现集落特征的最适培养温度为37℃。
【Abstract】 The characteristics of impression-slide cultures of the plague bacillus, Yersinia pestis, were that the nlarged bacil liwith dipolar staining curved naturally in "rope -end-like’’, "earthworm - like" or "knote-like" shaped chains which were short or long in appearance, on the agar contining 0.025% sodium sulphite and 1 % blood on hollow slides, over 4~6 hr cultivating. Moreover, part of chain groups twis-ted irregularly in a "bunch" or "fingerprint - like" or "strawhat - like" shaped colonies. These colonies dispersed in all field. The numbers of sacilli that formed chains and of chains that farmed primary colonies increased with the cultivating time and the inoculated quantities. The optimum temperature of cultivating was 37℃.
