含硒酵母的研究 Ⅰ、硒对酶母细胞生长的影响

【摘要】 啤酒酵母细胞从培养基中吸收硒(以亚硒酸钠形式供给)的数量和硒对酵母细胞生长的抑制作用,既和培养基中SeO3-2浓度有关,亦和细胞群体所处的生长阶段有关。培养基中SeO3-2浓度高于10-6M时,酵母细胞的生长就要受到抑制。在一定范围内,酵母细胞吸收硒的量,随着培养基中硒含量的增多而增加。在对数生长期,酵母细胞吸收硒的量最大。但在对数生长期给以10-3M亚硒酸钠,酵母细胞中有红色颗粒出现,可能是亚硒酸根还原而产生的元素硒的沉积。

【Abstract】 Selenium has long been known as an essential element of human body. A large tody of evidences of the preventive effect of selenium on cancer development and heart disturbance has been accumulated. While oral administration of inorganic selenium salt (sodium selenite) has been used in various districts in China as preventive means for endemic Ke-Shan disease (heart failure), selenium-yeast has been recommended in many countries. We started in 1982 the experiments of growing brewers yeast, Saccharomyces cerevisiae, in glucose-glycine yeast extract (GGY) medium containing various concentrations of sodium selenite with the aim of improving the method of producing yeast cells rich in selenium content.It was found that the inhibitory effect of sodium selenite on the population growth of yeast cells began to be detectable when the concentration of selenite was 10-6 M in GGY medium. Inhibition was enhanced with the increase of the selenite content of the medium. When the concentration of selenite raised to 10-3 M, practically no populaton growth occurred. While the population growth was inhibited, the cell size increased markedly and most of the cells carried buds. It seemed that selenium acted mainly to prevent the separation of the buds from the mother cells.The inhibitory effect of 10-3 M of sodium, selenite did not occur if the salt was added to culture which had grown for 16 hrs or longer, i. e., when it was at the later half of logarithmic growth and the following stationary phases. Reddish granules usually appeared in the cells if sodium selenite (10-3M) was added to logarithmic cultures. Apparently, the main constituent of these reddish granules was elementary selenium derived from the reduction of selenite (cp., Falcon, et. al., ).If sodium selenite was added 40 hrs after inoculation, i. e., to cultures which were at the early stationary phase with highest population density, no red granules could be seen in. the cells. However, the selenium content of these cells measured after 3 days of inoculation was still fairly high.
