HPD处理红光照射对离体培养人类正常细胞和肿瘤细胞的光敏效应

【摘要】 我们用细胞计数方法,测定HPD处理、红光照射对8株离体培养人类正常细胞和肿瘤细胞的光敏效应,正常细胞是短期培养的胎儿皮肤成纤维细胞及小儿包皮成纤维细胞,肿瘤细胞为建立细胞系,包括MGC80—3、SGC—7901、BEL—7402、Os—732、Detroit—6和HeLa。胎儿皮肤成纤维细胞和小儿包皮成纤维细胞,在单独5μg/mlHPD处理30分钟或单独用波长630nm红光100mw/cm~2照射10分钟,显示明显的毒性作用,而MGC80—3细胞生长增殖不受影响。但是,HPD处理红光照射明显增强对细胞的杀伤作用,5μg/mlHPD处理30分钟条件不变,MGC80—3细胞存活率随光照剂量(25mw/cm~25分钟、50mw/cm~25分钟及100mw/cm~210分钟)增加而下降。比较不同肿瘤细胞的HPD—红光光敏效应,5μg/ml HPD处理、照光50mw/cm~25分钟的细胞生长抑制率顺序为Os—732、MGC 80—3、Detroit—6.HeLa、BEL—7402和SGC—7901;而5μg/mlHPD处理、照光100mw/cm~210分钟的细胞光敏损伤效应依次为MGC 80—3、Os—732、SGC—7901、Detroit—6、HeLa及BEL—7402。最后,对正常细胞和肿瘤细胞的HPD—红光光敏效应问题进行了讨论。

【Abstract】 Photodynamic effects of HPD-red light in human mal ignantcell line ( MGC80-3, SGC-7901, BEL-7402, Os-732, Detroit-6 and HeLa) and normal cells (fibroblasts derived from human fetal skin and fibroblasts derived from child foreskin) were studied by cell counting. Cytotoxic effects were observed in human fetal fibroblasts and child foreskin fibroblasts but not in MGC80-3 cells when exposed to 5μg/ml HPD for 30 min. Or to 100mW/cm2 red light (630 nm in wave length) for 10 min. HPD treatment plus red light irradiation, however, increased significantly photoinactivation. When treated with the sane dose of 5μg/ml HPD for 30 min. The survive rate of MGC80-3 cells decreased in company with increase of irradiated doses of red light.In terms of the growth inhibition rates in culture, those cells treated with 5μg/ml HPD and irradiated with 50mW/cm2 red light for 5 min. Were Os-732, MGC80-3, BEL-7402, SGC-7901, HeLa and Detroit-6, respectively, while those cells treated with 5μg/ml HPD and irradiated with 100 mW/cm2 red light for 10 min. Were MGC80-3, Os-732, SGC-7901, BEL-7402, HeLa and Detroit -6, respectively.The results of HPD-red light photoinactivation on malignant and normal cells tested were discussed.
