HPD-红光对离体培养人胃癌MGC80-3细胞DNA、RNA和蛋白质前体掺入的影响
EFFECTS OF HPD PHOTOSENSITIZATION ON THE SYNTHESES OF DNA, RNA AND PROTEIN IN MGC80-3 CELLS IN VITRO
【摘要】 测定了HPD光敏作用对MGC80-3细胞DNA、RNA、蛋白质合成的影响,5μg/ml HPD作用30分钟,红光照射50mw/cm~25分钟,在细胞数量和活力没有改变的情况下,3H-胸腺嘧啶核苷掺入量明显减少,2小时后逐渐恢复,10小时和24小时超过对照,48和72小时又行减少,同时活细胞数量减少。3H-尿嘧啶核苷和3H-亮氨酸掺入始终没有因光敏作用而下降,前者24小时后掺入量还明显高于对照。文章对结果进行了讨论。
【Abstract】 The incorporation rate of 3H-TdR into MGC80-3 cells in vitrodecreased significantly from 5 min after treatment with HPD (5μg/mg for 30 min.) and red light (630nm, 50mw/cm2 for 5 min.), while the number and ciability of these cells remained at the same levels as in the control. The rate of 3H-TdR incorporation, however, increased gradually from 2 hr., and exceeded that of the control at 10 hr. and 24 hr. after HPD-red light treatment. However, at 48 hr. after treatment, the 3H-TdR incorporation rate, the cell unmber and the viability all decreased.Both RNA and protien synthesis did not decrease by HPD-red light treatment within 72 hr. of testing time. How-ever, the rate of 3H-uridine incorporation was much higher than that of the control.These results showed that 3H-TdR incorporation is a rapid, sensitive indicator for determining the effect of HPD phctosensitization on cancer cells in vitro. Since the rate of 3H-TdR incorporation decreased without loss of cell number and viability during the time of 2-24 hr. after HPD-red light treatment, it is suggested that HPD treated cells accumulated in G1 phase of the cell cycle and the transport function of the cell membranes was inhibited during this time.
