种传蚕豆真花叶病毒的研究
ON SEED-BORNE BROAD BEAN TRUE MOSAIC VIRUS IN CHINA
【摘要】 本文从生物学性状、理化特性、蛋白外壳及血清学特性等方面,比较系统地研究了从安徽蚕豆上分离到的一个种传分离物Bd。根据试验结果,参考国际上报道的文献,作者认为该分离物隶属于蚕豆真花叶病毒(BBTMV)。这是BBTMV 在中国的第一次报道。
【Abstract】 A sesd-borne virus isolate,Bd,was isolated from broad bean(Viciafaba)in Anhui,China.Its host range was very narrow,only broad beanand pea(Pisum sativum)could be infected.The dilution end-point wasdetermined to be 10-4 the thermal inactivation point was between 80 and85℃(10 minutes ),and the longevity in vitro at a room temperature(20-25℃)was 6 days.It was transmitted by sap inoculation and alsotransmitted by seed,but failed to be vectored by alfalfa aphids(Aphismedicaginis).The viral particles were purified from the infected leavesof broad bean by use of tetrachloride-PEG(MW.6000),followed by pel-leting them through a cushion of sucrose.The particles were isometric,25 nm in diameter,some of which were hollow.The UV absorption spe-ctrum was the typical one of the nucleoprotein,with A260/A280=1.83,and λmin=240nm,λmax=260nm.When sedimented in an analytical ultra-centrifuge,purified preparation of the isolate contained two centrifugalcomponents,having sedimentation coefficients(Sw.20)of 87.37 S and125.41 S.It had two eleetrophoretic forms in the electric field.The pro-tein shell of the virus consisted of two proteinsubunits of MW.c.44,000and 21,000.In agar double diffusion tests,the antiserum against Bd-iso-late showed good immunogenicity and specificity.ELISA method wassensitive to detect the antigenes both in the purified preparation and inthe sap of infected leaves.Based on the above results,it is primarilyconcluded that the Bd isolate belongs to broad bean true mosaic virusfirst reported by Quantz(1953).However this is the first report in de-tail about the occurrence of BBTMV in China.
