新疆哈密瓜上两种病毒的血清学特性

【摘要】 1982~1983年,作者等对从新疆哈密瓜上分离出的两种病毒西瓜花叶病毒2号(简称 WMV-2)和黄瓜花叶病毒(简称 CMV)进行了提纯,制备了抗血清。用部份提纯的 CMV 注射兔子,获得了1/512效价的抗血清。用制备的抗血清与 CMV-3(黄瓜株系)和 CMV-4(豇豆株系)进行了琼质双扩散,发现新疆哈密瓜上的 CMV 与 CMV-3和 CMV-4有一定差异,但与 CMV-3有一定相关。WMV-2提纯比较困难,因为在提纯过程中病毒极易聚集。用 PEG 沉淀法病毒重新悬浮困难,丢失较多,最后采用差速离心。在超速离心时加20%蔗糖垫。在两次差速离心后再经一次20~50%蔗糖梯度离心。得到了较好的提纯效果,制备了1/64效价的抗血清。用作者等制备的 WMV-2抗血清与美国、丹麦和日本的 WMV 抗血清进行了比较试验,证实新疆的 WMV-2与美国和丹麦的 WMV-2有相同的抗原性。用作者等制备的 WMV-2抗血清与哈密瓜病株的蔓、叶、花、果及种子进行血清反应,发现病株的蔓、叶、花、果各部均有大量病毒存在。

【Abstract】 The writers have made purification and prepared antiserum for two viruses(WMV-2,and CMV)isolated from saccharine melon in Xinjiang from 1982-1983. Cucumber mosaic virus(CMV)was purified by using clarification of expressed infected tobacco leaf sap in 0.5 M pH6.6 citrate buffer con- taining 0.1% 2-ME with 1/2 chloroform and two cycles of differential centrifugation,prepared for antiserum of CMV. Purification of water melon mosaic virus(WMV)is very difficult because of the aggregation of the virus particles during purification.After many experiments the following procedure was finally adopted.pumpkin leaf infected with WMV-2 was homogenized in 0.5M potassium phos- phate pH 8.5 containing 0.2% Na2SO3 and clarifieated by r-butanol(8 ml/ 100ml).The crude sap was collected and centrifuged at 8000 rpm for 20 min.The supernatant was removed and carbowax(PEG)was added at rate of 8g/100ml.After it was stirred for one hour,the misture was centrifuged at 8000 rpm for 20 rnin,and the pellets was resuspended in 0.02M potassium phosphate pH8.5.The resuspendant was concentrated by a cycle of differential centrifugation utilizing 20% sucrose cushion and futher purified by rate zonal sucrose dersity gradient cetrifugation, prepared for antiserum of WMV-2. Comparative experiment proved that WMV-2 of Xinjiang,China, USA and Dermak had homogeneity antigen and a large amounts of the viruses existed in stem,leaf,flower and fruit of the plant infected with WMV-2.
