治疗系统性红斑狼疮的新型免疫吸附剂的研究
INVESTIGATION OF A NEW IMMUNE ADSORBENT FORTREATING SYSTEMIC LUPUS ERYTHEMATOSUS
【摘要】 本文报道用火棉胶将氯化四氮唑兰〔2,2′,5,5′-tetraphenyl-3,3′-(3,3’-dimethoxyl-4,4′-diphenylene)ditetrazolium chloride〕(简称Blue tetrazolium,BT)与DNA形成的络合物固定到球形碳化树脂载体上的方法,制成用于临床治疗系统性红斑狼疮的新型免疫吸附剂。
【Abstract】 ABSTRACTThe article describes the immobilization of DNA and 2, 2’, 5, 5’-梩etra-phenyl-3, 3’- ( 3, 3’ - dimethoxy-4, 4’ -diphenylene ) ditetrazolium chloride ( blue tetrazolium, BT ) into the spherical beads of carbonized polymer adsorbent with collodion so that the formed complex may be clinically used to cure systemic lupus erythematosus ( SLE ) .With this adsorbent, a double amount of DNA is immobilized in the immune polymeric adsorbent as compared to that composed of active charcoal, DNA, and collodion.It has been reported that the amount of DNA immobilized with collodion into active charcoal is 0.4mg/g .charcoal and that immobilizfd in to the polymeric adsorbent is 0.92mg/g .resin.The percentage of DNA immobilized with collodion into active charcoal is 0.4mg/g. Charcoal and that immobilized in to the polymeric adsorbent is 0 .92mg/g .resin. The percentage of DNA immobilized in to the polymeric adsorbent is 98-98.5%.Factors affecting the amount of DNA immobilized in to the resin was also investigated .The polymeric adsorbent has good stability toward high tem-pe ature and acid. During hemoperfusion , the immobilized DNA did not liberate from the resin .The polymeric adsorbent show good compatibility with blood.2 hours after hemoperfusion, the platelet destruction is less than 20% in dogs Results indicate that in serum RlA of SLE patient the new polymeric adsorbent show specific seletive adsorption for DNA antibody and its immune complex,the rate of adsorption being>80% and that of the non-specific adsorption<10% .DNA and BT immobilied with the adsorbent show structures of single and double strands and may be used slinically with advantage.
