吉林省大豆属对根癌农杆菌致瘤反应及T-DNA转移的研究

【摘要】 本文报道了用根癌农杆菌的4个菌种感染吉林省大豆属2088份野生和栽培大豆基因型诱发致瘤的研究。筛选出100份适用于Ti质粒的受体材料;离体培养瘤的愈伤组织;建立了脱菌的悬浮细胞系;用纸上电泳测定证明胭脂碱存在,确定了T-DNA已经整合到大豆细胞中,经多次的继代培养不丢失,并能获得表达。

【Abstract】 Study OB tumor induced reaction of 2088 genotypes of Glycine in Jilin Province by infection, of four strains of Agrobacterium tumefariens was carried out. 100 genotypes have been screened, which can be used as a host system for Ti plasmid. In vitro culture of tumor tissues has been done. The suspended cell lines of bacteria-free tissue were set up. The presence of nopaline was analysed by using the paper electrophore-sis. After culturing several generations the T-DNA can still be detected for the presence of nopaline; dehydrogenase activity, so T-DNA might have been integrated into soybean genome.
