应用酶标记葡萄球菌A蛋白“ELISA”检测猪囊虫抗体的研究
APPLICATION OF ENZYMELABELLED SPA TO DETECTION OF CYSTICERCUS CELLULOSAE ANTIBODIES BY ELISA
【摘要】 作者进行了酶联免疫吸附试验(ELISA)检测猪囊虫抗体的研究,发现该法具有很高的敏感性和良好的特异性,少量假阳性主要由细颈囊尾蚴所致,可由细颈囊尾蚴液抑制而消除。以HRP-SPA代替HRP-兔抗猪γ球蛋白,对同一批猪血清进行了ELISA测定,发现两种结合物检测的结果一致,HRP-SPA完全可以代替HRP-兔抗猪γ球蛋白。SPA能与多种动物的IgG结合,而以与猪的IgG亲和力最强,同时它亦能与人的IgG结合。猪囊虫病为人猪共患的疾病,实验室只需准备HRP-SPA一种结合物,即可同时检测人和猪的囊虫病抗体,满足人医和兽医开展ELISA诊断囊虫病的需要。
【Abstract】 In our previous studies, ELISA method was used for the detection of antibodies of cysticercus ce-llulosae infection in pigs. The method was found to have extremely high sensitivity and relatively good specificity. There were some crossreactions between cysticercus cellulosae and cysticercus tenuicollis infections, but these false positive reactions could be inhibited by C. tenuicollis fluid, mixed with the swine sera. It was considered that ELISA might be a new and hopeful method. In this study, HRP-SPA was used to take the place of HRP-rabbit anti-pig Y globulin in the detection of the antibodies of C. cellulosae for the same batch of swine sera. The results obtained by HRP-SPA were the same as those by HRP-rabbit anti-pig Y globulin. It was thought that HRP-SPA might be a substitute for HRP-anti-pig globulin. SPA can be bound with IgG from many kinds of animals, among which pigs IgG shows the best affinity with SPA. Cysticercus cellulosae is a tissue parasite which can live in both pig and man. with such a conjugate as HRP-SPA in the laboratory, the cysticercus antibodies either from pig or from man can be detected. Therefore, HRP-SPA may be used in medical and veterinary practice for diagnosis of cysticercosis.
