【摘要】 采用 CM—Sephadex C—50,SP—Sephadex C—25离子交换柱层析和 SephadexG—50凝胶过滤三步分离程序,从东亚钳蝎毒中得到一种新的哺乳动物毒素。经低 pH 系统不连续聚丙烯酰胺凝胶园盘电泳,SDS—不连续聚丙烯酰胺凝胶板电泳及等电聚焦聚丙烯酰胺凝胶园盘电泳鉴定说明该毒素为电泳纯的蛋白质。用 SDS 电泳法测得其分子量为8,750道尔顿,等电聚焦电泳法测定 pH 为8.2。纯化成份的产率为2.5%。更多还原

【Abstract】 A new mammalian toxin,scorpion neurotoxin Ⅲ,was isolated and purified from the crude scorpion venom of Burthus martensii Karschi from Liaoning by ion-exchange chromatography on CM-Sephadex C-50 and SP-Sephadex C-25,gel filtration on Sephadex G-50.Its homogeneity was demonstrated by PH 4.5 polya- erylamide gel slab electrophoresis;isoelectric focusing electrophoresis and SDS-polya- crylamide gel slab electrophoresis.The molecular weight of the toxin,calaculated from measurement in SDS discontinuous polyaerylamide gel slab electrophoresis,are 8,750, and the isoelectrie point 8.2 by isoelectric focusing electrophoresis. To make the best of the available capacity,type with the designation C-25 should be used for separation of molecules below 10,000. Some experimental conditions were described.更多还原
