高效液相色谱分离7503光学异构体

【摘要】 本文将消旋的7503醇和光学纯的(+)或(-)的α-异丙基-4-氯苯乙酸反应,生成非对映异构体的酯衍生物,再用高效液相色谱分离。十分钟内可完成两种异构体含量的定量分析,其标准偏差小于0.4%。几百毫克量的制备可得到高效液相色谱纯的RS和RR或SS和SR两种组分。

【Abstract】 Enantiomers 7503 was first derivatized into diastereisomersby reacting with optically pure (+) or (-)The two diastereisomers were then separated completely within 10 min. on silica column by using hexane :ethyl acetate (500:7) or hexane containing 1% isopropyl ether as mobile phase. Isomers were prepared on a silica column (44×250mm) , with the injection amount of 400-600mg. The quantification was also made with a standard deviation less than±0.4%.
