血浆、尿液、脑脊液、腹水及生物制品中微量内毒素的定量检测法——鲎变形细胞裂解物改良基质显色法
QUANTITATIVE DETERMINATION OF ENDOTOXIN IN BLOOD,URINE,ASCITES,SPINAL FLUID AND BIOTIC AGENTS-MODIFIED LLT WITH CHROMOGENIC SUBSTRATE
【摘要】 本文介绍用改良的鲎试验基质显色法定量测定血浆、尿液、脑脊液、腹水和生物制品中的内毒素,并对测定原理和方法进行探讨。在最适条件下,本法可检出浓度≥50pg/ml 的内毒素。自身变异系数为3.5%,处理后变异系数为4.9%,回收率为50~60%。并且对革兰氏阴性细菌有特异性。热原测定结果与临床观察符合率达88.7%。采用国产 LAL 时,本法测定结果略优于日本基质显色法。临床初步检测和热原试验结果表明,改良的鲎试验基质显色法具有操作简便、省时、灵敏度高、重复性好和可以精确定量等优点,是目前诊断革兰氏阴性细菌感染,研究内毒素血症和检测热原的有效手段。
【Abstract】 Quantitative determination of endotoxin in the blood,urine,ascites,spinal fluid and biotic agents by using modified limulus lysate test(LLT)with chromogenic substrate, Boc-Leu-Gly-Arg-PNA was investigated.The assay was simple,accurate,time saving and reproducible,with a coeffeciency of variation of 3.5-4.9%,and a recovery rate of 50-60%.The sensitivity of the method was proved by its potency of detecting the endotoxin concentration as low as 50pg/ml.The method was an effective measure for tge examination of Gram-negative bacteria and pyrogens and the study of endotoxemia.
