【Abstract】 The causes of albuminous denaturation,such as pressure current velocity ,temperature, PH and so on,have initial been researched for the treatment albumin juice by ultrafiltration method.The theoretical analysis of the reasons on the albuminous denaturation has been carrihd out.It is suggested that the operational parameters of ultrafiltration treating albumin juice should be as the following:pressure 2～ 3 kg/cm~2,current velocity smaller than 50cm/sec.,temperature 10～20℃ ,PH5.8～7.2. 一、Introduction Ultafiltration is used to the albumin of biological product concentration and purification because the raw materials of biological product are biological activated,therefore,biological prodct not only fulfill requirement of purity quotient and concentration but also fulfill biological activated equirement.Ultrafiltration process would be controled in the fixed range of pressure,current velocity,temperature of solution and pH etc..We must avoid that the factors of physical and chemical cause albuminous chemical constitution changing in the operational process,or the biological product activated and useful value are reduced. Albuminous molecular weight is 69,000,ellipse,length is about 150A, diameter is about 38A.The principal action of aqueous albumin is keep osmotic pressure in the blood. 二、Technological process and equipments ( 一) Experiment process Experiment process is shown in Fig.1. (二 ) Equipment 1,External pressure control-type ultrafiltration instrument of polysulfone membrane. 2.Precision acidity test instrument (PHS-2) . 3.800 W electric furnace. 4.Stopwatch,mercury thermometer beaker,graduated cylinder etc.. 三、Test results ( 一) Current velocity The effects of current velocity on albumin denaturation is shown in Table 1. Experiment results have shown that current velocity could not be raised when ammoniasulfata density is higher concentration.Current velocity could be raised when ammoniasulfata density is lower concentration. (二)PH The experiment results are shown in Table 2. We must get under control pH. Making that pH get away albuminous equal electric point,we can avoid the phenomenon of albumin sedimentary. Experiment results have also shown that it integrated with the factors influncing the albumin denaturation.It is suggested that the operational parameters of ultrafiltration treating albumin juice should be as follows:pressur 2—3kg/cm~2, current velocity smaller than 50cm/sec.,temperature10-20℃, pH 5.8—7.2. We must control the ultrafiltration process so that the foam not appear.更多还原
