体外培养乳鼠心肌细胞的电生理学特性及与乳鼠成年鼠在位心脏的比较

【摘要】 作者使用常规的细胞内微电极技术,初步观察了由我院组胚教研室提供的培养乳鼠心室肌细胞的电生理特性,并与乳鼠、成年大鼠在位心脏相比较。本文结果与Schanne 0.F~3的报道相近似。用贴壁分离处理的心肌细胞静息电位较高,Vmax快。这一结果与Jourdon结果相似。可能是培养细胞中成纤维细胞的存在,某种原因降低了培养心肌细胞的电活动所致。作者在对比了培养心肌细胞、乳鼠、成年大鼠三者的电生理特性之后,对培养心肌细胞在电生理学研究中的应用,表明了粗浅的看法。

【Abstract】 The electrophysiological properties of cultured rat heart cells were investigated and compared with those of cells in intact hearts of newborn and adult rat. Cluster cell (500-1000 μm diameter) from newborn rat hearts (ventricles) were prepared and cultured for a period up to 7 days. Intracellular microelectrode recordings were made by using standard techniques. Cluster cell cultured under standard conditions had a low resting potential (-30 ± -7my) and the action potential had a slow rate of rise (10+ -3 v/s). Cluster cell cultured under differential attachment conditions had a high resting potential (-52±9mv)and the action potential had a fast rate of rise (132±10v/s). Reducing the amount of fibroblasts in the cluster cell by using the differential attachment technique caused the action potential parameter to become similar to those found iu non-cultured cells. Under stantard conditions, the electrical activity of cultured cluster cell from hearts of newborn rat differed from that recorded in infact heart of the newborn and adult rat.
