654—2对实验性呼吸窘迫综合征支气管肺泡灌洗液细胞的影响

【摘要】 用654-2防治油酸引起的狗呼吸窘迫综合征;分析观察了支气管肺泡灌洗液中吞噬细胞的数量、功能及超微结构等改变。结果发现,654-2能减轻油酸对肺泡巨噬细胞(AM)的损伤,保护AM的吞噬功能,减少因白细胞扣押于肺内而导致的肺损伤。

【Abstract】 The purpose of this study was to evaluate the mechanism of Oleic-Acid(OA)-induced lung injury and influence of Anisodamine (primarily extracting from a Chinese herb medicine) on the phagocytic cells,especially AM, by BAL.14 dogs were divided into two groups: Control group (n = 7, OA +NS) and experimental group (n = 7, OA + Anisodamine). The dogs were killed 90 minutes after the injection. AMs and PMNs were obtained from the lungs by saline BAL Phagocytic Percentage (PP) and Phagocytic Index (PI) of AM to C. albicans were determined. Compared with Anisodamine group, control group’s PP (4 . 04 ± 0. 67%) vs 10.80±2.35%) and(0.06±0.01 vs 0.28 ± 0.06) were decreased (P<0.05 or 0. 01); the surfaces of AMs were smooth and the cytoplasm of AMs included many lipid particles under the transmission electron microscope and scanning electron microscope respectively; there was marked increase in PMN by evaluation of BAL (28.2 ± 5.6 × 103/ml vs 7.7±2.3× 103/ml) and the lung tissue sections stained with HE(105±13vs 61.02±6.08/10 fields).These findings suggest that:1) AMs and PMNs play an important role in mechanism of ARDS. 2) The eflcct of Anisodamine on the RDS may have relation to its influence on AM phagocytic capability.
