EFFECT OF ANISODAMINE ON PULMONARY FUNCTIONOF DISAGGREGATING PLATELET MICROAGGREGATES IN CANINE ENDOTOXEMIA
【Abstract】 This work studied the effect of Anisodamine (primarily extracting form a Chinese herbal medicine) on one of the pulmonary non-respiratory functions: disaggregating platelet microaggregates (MA) in endotoxemic dogs. The results showed that the platelet aggregating rates (PAR) in left ventricular blood (LVB) were significantly lower than that in right atrial blood (RAB) in normal anesthetized dogs (p<0.05). After Anisodamine injection, the PAR of experimental animals (N = 11) decreased (p<0.05); whereas the controls (N= 11) remained unchanged. 60 minutes after dead E coli injection, there was no longer any difference of PAR between RAB and LVB in the controls, and remained as such later, but this difference of Anisodamine-treated dogs existed throughout the experimental period. Also, Anisodamine minimized the early increades of PAR and decreases of blood pressure and PaO2 induced by endotoxin. These data suggest that 1) Anisodamine may protect pulmonary disaggregating function of endotoxemic dogs; and 2) Anisodamine may disaggregate MA in peripheral blood and attenuate the early effect of endotoxin. We conclude that the efficacy of Anisodamine in clinical treatment probably relates to its effect on the non-respiratory functions of the lungs.
