血卟啉衍生物加光照引起Ehrlich腹水癌细胞的化学发光和膜脂过氧化作用
CHEMILUMINESCENCE AND MEMBRANE LIPID PEROXIDATION INDUCED BY HEMATOPORPHYRIN DERIVATIVE PLUS LIGHT OF EHRLICH ASCITES TUMOR CELLS
【摘要】 在HPD和光照处理后,细胞的化学发光计数显著高于单用HPD或单用光照以及未经任何处理的细胞化学发光。缺氧和单态氧淬灭剂(β-胡萝卜素和叠氮钠)能降低细胞的化学发光。与各种对照组相比,经HPD加光照处理的细胞的膜脂过氧化物量显著增加,膜的流动性相应地下降。HPD在肿瘤的光辐射治疗中的作用机制,在于单态氧(或自由基)启动的膜脂过氧化作用,导致膜的生物物理性质变化,进一步使细胞的结构和功能改变。
【Abstract】 The data of effects of HPD plus light on chemiluminescence, membrane lipid peroxidation and membrane fluidity of Ehrlich ascites tumor cells were presented.The chemiluminescence count of Ehrlich ascites tumor cells treated with HPD plus light was much higher than that treated with HPD or light alone and that without any treatment. Anoxia, and quenchers of singlet oxygen(β-carotene and sodium azide)inhibited chemiluminescence of cells. Compared with the other cell groups mentioned above, the amount of membrane lipid peroxide of cell treated with HPD plus light significantly increased. Corresponding to the increase in lipid peroxid, membrane fluidity of cells decreased. Based on these results obtained, authors suggested that the mechanism of action of HPD in conjunction with cancer photoradiation therapy lie in membrane lipid peroxidation initiated by singlet oxygen (or radicals), which led to changes in biophysical property of membrane and further to alteration of structure and function of cell.
