HpD—激光对动物瘤体的细胞化学影响(选用细胞色素氧化酶、琥珀酸脱氢酶、酸性磷酸酶显示法)

【摘要】 本文为研究HpD-激光破坏恶性肿瘤的机理,采用细胞化学方法观察了小白鼠肿瘤中细胞色素氧化酶、琥珀酸脱氢酶和酸性磷酸酶的活性。在HpD-激光处理瘤细胞后12小时细胞色素氧化酶活性明显降低,24小时即消失。然而,琥珀酸脱氢酶和酸性磷酸酶无明显变化。结果表明,HpD-激光破坏恶性肿瘤的原因之一,乃是抑制了瘤细胞内的细胞色素氧化酶活性,进而阻断了呼吸链。
【Abstract】 Enzyme activity of cytochrome oxidase, succinic dehydrogenase and acid pho-sphatase was observed by using cytochemistry method in rat tumors in order to study the mechanism of Hematoporphyrin derivative-photodynamic therapy (HpD-PdT) to destroy malignant tumors.Enzyme activity of cytochrome oxidase decreased significantly at 12 hours and disappeared at 24 hours after exposure of tumor cells to HpD-PdT technique.However, the enzyme activity of both succinic dehydrogenase and acid phosphatase was not obviously changed.It shows that destruction of malignant tumors by HpD-PdT might be due to inhibition of cytochrome oxidase and interruption of respiratory chain in the tumor cells.
