新疆大麦条纹花叶病毒的研究——Ⅰ.病毒株系类型的鉴定及核酸的体外翻译
STUDY OF BARLEY STRIPE MOSAIC VIRUS ( BSMV) XINJIANG ISOLATE Ⅰ.IDENTIFICATION AND TRANSLATION OF VIRAL RNA COMPONENTS
【摘要】 我国大麦条纹花叶病毒(BSMV)新疆分离物的RNA,在50%甲酰胺6%甲醛中,50℃处理15分钟,使RNA分子链呈真正的均一状态后,在含甲醛的琼脂糖凝胶上电泳,核酸变性电泳结果显示该病毒为三组分RNA株系,编号为RNA1,RNA2,RNA3,分子量分别为1.40—1.44×10?,1.20—1.28×100,1.06—1.09×106。经制备凝胶电泳分离出的各RNA组分在兔网织红细胞溶胞液的无细胞体系中,RNA1的翻译产物为120K的蛋白,RNA2翻译产物主要为25K和一个85K蛋白,其中25K蛋白由于与天然BSMV外壳蛋白共电泳,该蛋白中没有甲硫氨酸,以及能被BSMV抗血清沉淀,而被确定为BSMV外壳蛋白。RNA3的翻译产物为75K蛋白。
【Abstract】 Total RNA of Barley Stripe Mosaic Virus ( BSMV ) isolated from Xinjiang was heated at 50℃ for 15 min.in 50% formamide and 6% formaldehyde and electrophoresed in agarose: formaldehyde gel. Three RNA components RNA1, RNA2, and RNA3 with M.W.of 1.4-1.44× 106, 1.2-1.28×106 and 1.06-1.09×106 respectively were found.The translation products of each RNA component in rabbit reticulocyte cell-free system were investigated. RNA1 directs a 120K. polypeptide. RNA2 directs 25K and 85K; RNA3 a 75K polypeptideThe 25K polypeptide directed by RNA2 was recognized as the viral coat protein based on the evidence:1.The in vitro translated 25K protein.completely comigrated with the authentic coat protein, as they were coelectrophoresed.2.When85S-methione was used as the labeled amino acid initro system, 25K disappered, which coincides with the fact that there is no methio-nine residue in the authentic protein of BSMV Xinjiang isolate.3.When the translation products were precipitated by the rabbit antiserum against BSMV, only 25K polypeptide specifically appeared on the gel.
