蚕豆真花叶病毒的鉴定——Ⅰ 生物学性状、提纯及血清学特性的研究
CHARACTERIZATION OF BROAD BEAN TRUE MOSAIC VIRUS IN CHINA
【摘要】 从安徽蚕豆上分离到一个种传病毒分离物Bd,仅能侵染蚕豆和豌豆,稀释终点(DEP)为10-4,致死温度(TIP)80~85℃(10分钟),体外存活期(LIV)6天(20~25℃室温);可经汁液和种子传播,但苜蓿蚜(Aphis medicaginis Koch)不能传毒。用四氯化碳——PEG(MW.6000)法,结合蔗糖垫差速离心,可获高度纯化的病毒制备物。病毒粒子等轴对称,直径约25nm,部分粒子被染料渗透而中空。用纯化的Bd分离物制备的抗血清表现高度的专一性,在琼脂双扩散试验中,Bd分离物与其抗血清只形成一条沉淀带;应用酶联免疫吸附试验(ELISA)成功地检出了该分离物,并获得较高的灵敏度。据此,作者初步认为Bd分离物类似于Quantz等报道的蚕豆真花叶病毒(Broad Bean True Mosaic Virus)。本文是蚕豆真花叶病毒在我国的第一次报道。
【Abstract】 A seed-borne virus isolate, Bd, was isolated from broad bean (Vicia faba) in Anhui. Its host range was very narrow, only broad bean and pea (Pisum sativum) could be infected. The properties in sap, dilution end-point was 10-4, thermal inactivation point was 80—85℃ (10 minutes), and longevity in vitro at a room temperature of 20—25℃, was 6 days. It was transmissible by sap inoculation and by seed, but failed to be vectored by alfalfa aphids (Aphis medicaginis). The viral particles could be purified from the infected leaves of broad bean by use of tetrachloride-PEG (MW. 6000) method, followed by pelleting them through a cushion of sucrose and then a virus preparation with high purity was obtained. The particles were isometric, c.25 nm in diameter, some of which were hollow. In agar double difusion tests, the antiserum against Bd-isolate showed good immunogenicity and specificity. ELISA has successfully detected the antigenes both in the purified preparation and in the sap of infected leaves. Based on the above results, and also on the information obtained from the study about the UV-spectrum,sedimentation properties, electrophoretic forms and the capcid proteins of the isolate (in publishing), it is primarily concluded, therefore, that the Bd-isolate belongs to broad bean true mosaic virus reported by Quantz (1953). This is the first report on BBTMV in China.
