脂质体携带抗癌药物对动物实验肿瘤的研究

【Abstract】 This study is to investigate the distribution,toxicity and effectiveness of liposome-encapulated-antitumor agents (L-drugs), such as cytoxan (L-CTX), vincristine (L-VCR) and 5-fluorouracil (L-Fu), as compared to free CTX, VOR and 5-Fu. The resultsare as follows: (1) The distribution of L-99mTc and free 99mTc: free 99mTc appeared mainly inthe stomach, while L-99mTc was in liver and spleen. (2) The acute LD50of L-VCR and L-CTX compared to free CTX and VCR arequite different. Free VCR vs L-VCR is 1.09mg/kg: 1.70mg/kg and free CTX vsL-CTX is 316.5mg/kg: 448.3mg/kg(P<0.01). (3) The effectiveness of L-drugs in tumor-bearing animal was compared with freedrugs: L-CTX in low dose (10mg/kg) had better antitumor effect than free CTX, but L-VCR and free VCR were about the same. It is very interesting that if free VCR wasused to treat the mice with ascitic type of liver cancer, there were only 2 mice living over30 days, a medium survival of 21.3 days (the control group was 13.9 days), while inL-VCR group there were 5 mice living over 30 days, a medinm survival of 34.2 days(P<0.05).
