阿魏酸钠抗血小板聚集作用机理研究——对TXA2/PGI2平衡的影响

【摘要】 大鼠口服 SF(600mg/kg)后2小时,以胶原诱导的血小板聚集性及血小板TXA2样物质的活性均受显著抑制,抑制率分别为46%及47%,对动脉壁PGI2活性则无明显影响;SF体外给药有桔抗血小板TXA2样物质活性(0.25~1.0mg/ml)和增细PGI2活性的作用(0.5mg/ml)。可见SF 通过多种途径影响 TXA2/PGI2平衡,对血栓性疾病防治似有裨益。

【Abstract】 Male rats were treated with sodium ferulate (SF, 600mg/kg po) or aspirin (300mg/kg po) and were sacrificed 2 hours later. Platelet aggregation induced by collagen Was measured. TXA2 activity was estimated by measuring the contraction of rabbit thoracic aorta and PGI2 activity by measuring inhibition of rabbit platelet aggregation induced by ADP. The platelet aggregation and TXA2 activity were inhibited by 46 and 47%, respectively with SF and by 47 and 96%, respectively with aspirin. At the same time, the carotid arterial PCI2 release remained unchanged with SF and obviously suppressed by 91% with aspirin. In in vitro studies, SF showed significant antagonistic effect on pooled rat platelet TXA2 activity induced by collagen. SF at the concentration of 0.5mg/ml exhibited obvious potentiating effect on inhibition of platelet aggregation by either rat carotid arterial segment incubates or sodium salt of PGI2 These results indicate that SF may be effective in treating thrombosis.
