【Abstract】 The hepatitis B surface antigen (HBsAg) cDNA, containing the complete coding sequence, was ligated to the Lac promotor in a plasinid at the right orientation by ECoRI linker. The chimeric DNA was then used to transform E. Coli RRI. The expression of HBsAg was detected by 35S-methionine incorporation. The result showed the expressed HBsAg has a molecular weight of 24000, and is very close to the molecular weight of the native HBsAg.
