葡萄糖氧化酶电极的研制及其应用的初步试验
A PRELIMINARY STUDY ON THE PREPARATION AND APPLICATION OF COLLAGEN-----GLUCOSE OXIDASE ELECTRODE
【摘要】 本文介绍了以胶原为载体用包埋交联法制备葡萄糖氧化酶膜的方法和条件。在选用的酶量范围内,每10g胶原与100mg葡萄糖氧化酶包埋制得的酶膜响应值最大,用0.1%戊二醛交联30秒制备的酶膜稳定性较好,响应值较大。与溶氧电极组装成的电极其测定的最适温度为37℃,最适pH为5~7。80mg/10g胶原的酶膜响应时间为10~30秒,平衡时间为3~7分。每更换一次缓冲液,进量为0.1ml时,可连续进样十余次,重演性好,宜用于连续测定。测定红霉素种子罐培养液和60小时前的发酵液糖含量均比用Fehling法的略低,但规律性及波动情况大致相同。我们初步认为这种电极有可能被用于测定红霉素发酵液的糖含量。这种用胶原酶膜与溶氧电极组装的葡萄糖电极以测定氧消耗的电流型电极尚未见报道。
【Abstract】 An enzyme electrode consisting of an immobilized collagenglucose oxidase membrane and oxygen electrode wes prepared for the estimation of glucose in fermentation broth.The collagen-glucose oxidase membraneshave been prepared by casting the collagen-glucose ozidase dispersion on a teflon disk. After drying, the membranes were ciosslinked in glutaraldehyde solution at various. times and thoroughly washed.It was found that the membranes prepared with glucose oxidase concentration of 80 mg per 10 grams collagen gel and cross-linked with 0.1% glutaraldehyde for 30 seconds were quite stable under our experimental conditions. The enzyme electrode with thus prepared membrane was quite sensitive and reproducible. In the determination of a 0.1 ml glucose sample (2.5%)the response time was only 10-30 seconds and the resposevalue was large.Ten glucose sample solutions (0. 1ml, 2.5%)can be continuously determined within one batch buffer solution.The optimum temperature was 37℃ and the optimum pH was 5 to 7.In the determination of the glucose content in the Erythromycin seed cultures and the 0 to 60 hours Fiythromycin fermentation broths, good comparable results were obtained between the conventional Fehling method and the enzyme electrode method.
