青霉素球状菌株发酵工艺的研究——Ⅳ.铁的影响
STUDY ON FERMENTATION PROCESS OF PELLET FORM OF Penicillin chrysogenum Ⅳ. INFLUENCE OF IRON
【摘要】 <正> 铁对青霉素发酵的影响,很早就引起注意。近年来,青霉素发酵单位产量逐步上升,铁的影响在工业生产上更加被重视。为减少铁对青霉素发酵的干扰,大都采用三种方法:使用不锈钢材料制备生物反应器及其附属设备以减少铁的污染;或以惰性涂料涂布在碳钢表面,以减少铁的腐蚀溶解;或增大生物反应器容积,以增大培养液体积与铁的接触面积的比值。近年来,曾通过菌株选育获得对铁敏感性较低的新菌株,以降低工业生产中铁对青霉素发酵的干扰。
【Abstract】 Experimental results showed that, there was no influence of iron upon the growthand development of pellet form of Penicillium chrysogenum but it depressed thebiosynthesis of penicillin. The depressing intensity was increased as the concentrationof iron increased in the fermentation medium. When the iron concentration was over 50γ/ml in the inoculation stage ofindustrial fermentation process, the fermentation medium may be contaminated, andthe yield of penicillin depressed. The maximum influence intensity of iron uponbiosynthesis of penicillin may be obtained just around the 96th hour of fermentation.Iron may be concentrated into fungal mycelium, and released into broth during myce-lium autolyzation. The intensity of depression of penicillin biosynthesis by insoluble ironsalts wasweaker than soluble iron salts. But both of them could be transformed mutually.Sterilization process may promote the transformation. Inorganic chelating agents of iron may depress the penicillin biosynthesis too.Consequently, it did not improve the yield of penicillin, which had been contami-nated by iron.
