单克隆抗体对银屑病初发损害内浸润细胞的免疫组织化学表现型的初步鉴定

【摘要】 初发银屑病的细胞浸润成分与银屑病的发病机制密切相关,本研究应用间接免疫过氧化酶技术对四份初发银屑病损害标本,应用抗T细胞、T细胞亚类、轻键、单核细胞和Ia样抗原的单克隆抗体进行单个核浸润细胞成份的鉴定。结果表明,初发性银屑病损害内主要系Lyt 2~+、Lyt 3~+、Leu 4~+、Leu 3~+和Ia~+细胞浸润,浸润的角化不全部的表皮及真皮浅层最为明显并向周围递减。另外,Leu 2~+细胞在表皮内相对明显增多,这一现象在角化增厚部表皮为明显,这一现象也可能适用于他其上皮的病变。本研究尚证实银屑病初发皮损的角化不全区有特异性免疫球蛋白的沉积,这一变化已证明晚于表皮病变。

【Abstract】 The cell infiltration is one of the basic pathologic changes of psoriasis. Possibly it is close related to the pathogenesis of psoriasis. Biopsies of initial psoriasis skin lesions were examined with various monoclonal antibodies against T lymphocyte. T subset, B lymphocyte, monocyte Ia antigen K andλ chains using immunoperoxidase technique. It was shown that the majority infiltrating cells in dermis of initial psoriatic skin lessions were Lyt2~+, Lyt3~+, Leu4~+, Leu3~+ and Ia~+cells. Mostly. they were in the upper part of dermis beneath the parakeratosis epidermis. A decreasing cell infiltraiion was found in the dermis baneath orthokertosis area. However there was a significant high proportion of Leu2~+ cells in the epidermis. It was more Pronounced in the ortho keratosis epidemis than that in the parakeratosis epidermis. The cell infiltrates in the adjecnt normal skin did not differ from that of normal skin taken from psoriatic patients and normal donorsi This phenomenon could be applied for other epithelial pathologic changes.
