【摘要】 从甘肃岷县当归挥发油中,分离出一淡黄色油性物质,鉴定为藁本内酯,实验证明有明显的平喘作用。给豚鼠ip 0.14 ml/kg能缓解组织胺与乙酰胆碱的致喘反应,其效力约与氨茶碱50 mg/kg相仿。在豚鼠肺溢流实验,ⅳ藁本内酯0.08 ml/kg能对抗组织胺2～10 μg/kg所引起的支气管收缩。藁本内酯不仅对豚鼠离体气管条有松弛作用,而且对乙酰胆碱、组织胺以及氯化钡引起的气管平滑肌痉挛收缩,有明显的解痉作用。上述作用不能被心得安所阻断。藁本内酯对豚鼠肺、肠组织内cAMP及cGMP的含量均无明显影响。更多还原

【Abstract】 A component, ligustilide, was isolated from the essential oil of Danggui (Angelica sinensis Oliv. Diels) cultivated in Min Xian County, Gansu Province, and accounted for 0.5% of the weight of the original herb. It was shown to have remarkable antiasthmatie effect and spasmolytie action by a series of experiments. When injected into guinea pigs intraperitoneally at a dose of 0.14ml/kg, the asthmatic reaction induced by acetylcholine and histamine was immediately impeded with a potency approximately equal to the action of aminophylline (50mg/kg). Lung overflow experiment showed that an intravenous injection of ligustilide (0.08 ml/kg) into the anesthetic guinea pig caused complete or partial block of the reaction of histamine at the dose of 2～10μg/kg. Ligustilide also exhibited antispasmodic effect on isolated trachea strip of the guinea pig contracted by acetylcholine, histamine and barium chloride, and a relaxation effect on trachea strip under normal tension. Addition of propranolol did not affect those actions. The contents of cAMP and cGMP in the pulmonary and intestinal tissues of the guinea pig were not altered by treatment with ligustilide.更多还原
