三基单倍体水稻胚性细胞团的诱导及植株分化的研究

【摘要】 稻属种间杂种(Oryza sativa×O. latifolia)F1(三基单倍体)的幼穗,在HE培养基中,首先发生愈伤组织,两个月后,在愈伤组织上产生乳白色颗粒状、易分散的胚性细胞团。但每个胚状体本身是坚实的。试验表明,以HE或MS为继代培养基,以MS为分化培养基,细胞团分化出苗的效果最好,成苗率可达80%以上。如每月转入新鲜的继代培养基,则细胞团的繁殖与分化的能力可长期保持在较高的水平上。

【Abstract】 The immature panicles of the first offspring of a specific hybrid of Oryza sativa×O. latifolia, a trihaploid rice, were cultured on HE medium containing 2, 4-D 2mg/l, NAA 2 mg/l and kinetin 3 mg/l. Callus was first induced in culture and then embryogenic cell cluster milk white in color could be easily separated. But the embryoids were rather firm, occurring from the callus about two months after inoculation. Various developmental stages of embryoids such as globular-shape, heart-shape, scutellum-shape and mature embryoids were found within an embryogenic cell cluster.The media of both subculture and differentiation had a great effect on the differentiation of embrygenic cell cluster. It was shown from the experiment that higher frequency of plantlet (more than 80%) could be got when HE and MS were used as subculure media and MS was used as differentiate medium.The ability of embryogenic cell cluster to proliferate and differentiate attained its maximum when embryogenic cluster was cultured on the subculture medium 30-60 days after being translated. However, when the subculturing interval prolonged to more than two months, then the longer the subculturing time was, the weaker the abilities to proliferate and differentiate of the embryogenic cell cluster were; and eventually the embryogenic cell cluster would lose its ability to proliferate and differentiate if the embryogenic cell cluster was subcultured for 4-5 months without translating to fresh medium. Nevertheless, the embryogenic cell cluster could maintain its ability of proliferation and differentiation to a high level and last for a long time if the embryogenic cell cluster was subcultured monthly.
