L-脯氨酸生物合成条件的研究

【摘要】 作者以谷氨酸棒状杆菌(Corynebacterium glutamicum)№.11为试验菌,进行了L-脯氨酸生物合成的研究。观察了葡萄糖、废糖蜜、氯化铵、硫酸铵、硫酸镁、氯化镁、生物素、豆饼水解液、乙醇以及pH等因素对L-脯氨酸生物合成的影响,并找到了此菌在生物合成脯氨酸的最佳条件。 我们以廉价废糖蜜为原料直接发酵生产L-脯氨酸。采用豆饼水解液省去价格昂贵试剂肉胨及硫胺素盐酸盐。同时以中途流加尿素替代碳酸钙,为L-脯氨酸生产工业化提供条件。L-脯氨酸的最高产量水平为35mg/ml,转化率约25%。

【Abstract】 On purpose to study the biosynthesis of L-proline by strain Corynebacterium glutanicum No. 11, the effect of glucose, molasses, ammonium chloride, ammonium sulfate, magnesium sulfate, magnesium chloride, biotin, hydrolysate cake meal, alcohol and pH on the biosynthesis of L-proline was observed and the best conditions of L-proline’s biosynthesis for this strain were determined. This paper also reports that we used cheaper molasaes to ferment L-proline and the hydrolysate of soybean cake meal to replace the more expensive reagents of peptone and thiamine hydroohloride. The urea was added directly and continuously to replace calcium carbonate. The results of this method offer favorable conditions for amino acid industry. The highest level of amino acid is 35 mg/ml. The conversion ratio is 25%.
