运铁蛋白及其抗血清的制备和癌细胞株运铁蛋白受体的检测

【摘要】 本文介绍用利凡诺(Rivanol)沉淀、硫酸铵盐析及DEAE-SephadexA50层析法提纯人血清运铁蛋白(T?),并用它免疫家兔,制备兔抗人运铁蛋白单价特异抗血清。采用间接免疫酶标法检测人体培养细胞株运铁蛋白受体。证明人宫颈癌培养细胞(Hela细胞)及五株人肝癌培养细胞(BEL-7402、BEL-7405、SMMC-7721、QGY-7703及Mahlavn)的细胞膜上均具有丰富的运铁蛋白受体。

【Abstract】 Preparation of transferrin from human serum was described. In brief it was precipitated by Rivanol and then fractionated by ammonium sulfate and DEAE-Sephadex A-50. The final product was essentially pure,, with contaminant of other proteins less then 4% on polyaorylamide gel eleotrophoresis.Babbit antitransferrin antiserum was produced. Transferrin receptor was detected by indirect peroxidase-linked immunohistochemistry technique. 6 tumor cell strains were examined (Hela, liver cancer BEL-7402, BEL-7405, SMMG-7721, QGY-7703 and Mahlavn), all exhibiting plenty amount of transferrin receptor on their cell surfaces.
