带SRS肿瘤小鼠脾细胞对体外淋巴细胞增生反应的抑制作用
THE INHIBITORY EFFECT OF SPLEEN CELLS FROM MICE BEARING SRS-TUMOR ON THE PROLIFERATIVE RESPONSES OF LYMPHOCYTE IN VITRO
【摘要】 本文报道接种SRS肿瘤后,带瘤小鼠的脾脏细胞具有非特异性抑制体外同种异型细胞(MLR)和淋巴细胞对致分裂原刺激的增生反应。这种抑制效应在肿瘤接种后3天即可观察到,其中以粘附细胞的抑制作用为明显,从带瘤10天、20天、33天小鼠脾脏组分出来的粘附细胞,对刀豆素A(ConA),大肠杆菌脂多糖(LPS)诱发淋巴细胞增生反应的抑制率均可达90%以上。用60钴2500rad照射或用抗Thy-1血清和补体处理这种脾脏粘附细胞,只能部分地消除它的抑制作用,提示很可能是通过粘附细胞中的巨噬细胞发挥作用,但也不能排除T细胞在其中也起一定的作用。
【Abstract】 A marked inhibitory effect on in vitro proliferative response of lymphocyte to mitogens (Con A, PHA, LPS) and allogeneio cells were induced by the spleen cells from SES-tumor-bearing mice. Such inhibitory effect began to appear on the 3rd day after inoculation. It was also shown that the adherent spleen cells had stronger inhibitory effect than those induced by the same number of total spleen cells from SRS-bearing mice. The adherent cells fractionated from lOd, 20d and 33d tumor-bearing mouse spleen all showed an extremely marked inhibition on mitogenic response of lymphooyte up to 90%. Irradiation (2,500 rads) of adherent spleen cells resulted in partial abrogation of their inhibitory effect. Furthermore, similar result was observed when cells were treated with anti-thy-1 serum.Our data indicated that, besides radioresistant cells (probably macrophages), the radiosensitive T subset might be involved in the inhibitory effect of adherent cella from spleen of SRS-bearing mice.
