用原位电泳技术测定艾氏腹水癌细胞膜的流动性
THE MEASUREMENT OF MOBILITY OF EHRLICH ASCITIC CARCINOMA CELL MEMBRANE COMPONENTS WITH IN SITU ELECTROPHORESIS
【摘要】 细胞膜组分(膜蛋白、膜脂等)分子能沿膜进行侧向扩散运动,研究这种运动对了解细胞的功能有重要意义。原位电泳(In Situ Electrophoresis)是近年发展起来的测量细胞膜组分侧向扩散的新技术之一。我们用这种技术研究了小鼠艾氏腹水癌细胞膜ConA(Concanavalin A)受体的侧向扩散,并测得在22℃时,其侧向扩散系数为3.2×10-10cm2/see。实验结果提示,侧向扩散系数具有较大离散性可能是癌细胞的特征之一。
【Abstract】 Cell membrane components(proteins,lipids etc.)can diffuse along the membraneplane.The study of such movements is useful in understanding cell function.In situelectrophoresis is one of the new techniques applicable to the measurement of thelateral diffusion of the molecules.By this technique,the lateral diffusion of Concan-avalin A receptors in the membrane of Ehrlich ascitic carcinoma(EAC) cells,werestudied.The diffusion coefficient D=3.2×10-10cm2/sec at 22℃,a larger dispersionof the diffusion coefficient was noticed.It is suggested that this is probably one of thecharacteristics of cancer cells.
